Description: The lymphocyte activation gene-3 (LAG3) is a member of the immunoglobulin superfamily and binds MHC class II with high affinity (1), negatively regulating T-cell function and homeostasis (2). It is expressed in B, T, and NK cells, monocytes, and dendritic cells (3), and acts to regulate T cell expansion (4). LAG3 is also an important immune checkpoint protein, with anti-LAG3 antibodies activating T effector cells and affecting regulatory T cell functions. Furthermore LAG3 appears to act in a synergistic fashion with PD-1/PD-L1, suggesting that a dual antibody approach may prove useful in cancer immunotherapy (5). |

ProSci | 0.1 mg | EUR 594.26 |
Description: VISTA Antibody: VISTA/B7-H5/platelet receptor Gi24 is a single-pass type I membrane protein located at the cell surface. It is an immunoregulatory receptor which can inhibit T-cell response and may promote differentiation of embryonic stem cells, by inhibiting the BMP4 signaling pathway. The protein can be cleaved by MMP14, and stimulate MMP14-mediated MMP2 activation. |

NSJ Bioreagents | 100ug | EUR 349.3 |
Description: VISTA / GI24 is a transmembrane protein expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces. On ESC s, Gi24 appears to positively interact with BMP-4, potentiating BMP signaling and the transition from an undifferentiated to a differentiated state. On tumor cells, Gi24 both promotes MT1-MMP expression and activity and serves as a substrate for MT1-MMP. This increases the potential for cell motility. Mature human Gi24 contains a 162aa extracellular region with one V-type Ig-like domain and a 96aa cytoplasmic domain. Human Gi24 undergoes proteolytic cleavage by MT1-MMP, generating a soluble 30kDa extracellular fragment plus a 25-30kDa membrane-bound fragment. VISTA is a negative checkpoint regulator and is expressed on myeloid cells, T-cells and human TILs (tumor infiltrating lymphocytes) on MDSCs (myeloid-derived suppressor cells) in the TME (tumor microenvironment). It is very likely both a ligand and receptor and is a promising target for cancer immunotherapy. |

Description: VISTA-IN-2 (Compound 1) is an inhibitor of V-domain Ig suppressor of T-cell activation (VISTA). VISTA-IN-2 induces VISTA degradation in cells through an autophagic mechanism. VISTA-IN-2 rescues VISTA-mediated immunosuppression, and enhances antitumor activity of immune cells. VISTA-IN-2 also activates the antitumor immunity and inhibits tumor growth in a CT26 mouse model[1]. |
